Nowadays, Lab On Chips (LOCs) require the development of new technologies in order to integrate complex fluidics, sensors, actuators... Such integration requires overcoming both technological bottlenecks and an increase in term of production cost. We propose a technique to manufacture reusable and complex LOCs made up of SU-8 resist for the fluidic structure, of glass for the hard packaging and are compatible with the integration of thick electrodes. The method is based on the combination of two bonding technologies, both based on a wafer bonder. The first one consists in the bonding of a thin photosensitive SU-8 dry film, which is similar to lamination. The second one is the standard bonding technique which uses a hard substrate covered by a SU-8 layer. The LOC that can be obtained thanks to the combination of these two methods are transparent, can include 3D microfluidic structures, and thick electrodes. Moreover these LOC are reusable, packaged and ready to use. In order to validate the concept we designed a LOC devoted to cell arraying, using dielectrophoresis, as well as to cell electroporation.

Lab-on-a-chip systems offer a versatile environment in which low numbers of cells and molecules can be manipulated, captured, detected and analysed. We describe here a microfluidic device that allows the isolation, electroporation and lysis of single cells. A431 human epithelial carcinoma cells, expressing a green fluorescent protein-labelled actin, were trapped by dielectrophoresis within an i...

Dielectrophoresis-assisted 3D nanoelectroporation for non-viral cell transfection in adoptive immunotherapy.

Current transfection technologies lead to significant inter-clonal variations. Previously we introduced a unique electrotransfection technology, Nanochannel-Electroporation (NEP), which can precisely and benignly transfect small cell populations (~100-200 cells) with single-cell resolution. Here we report on the development of a novel 3D NEP system for large scale transfection. A properly-engin...

Controllable in-situ cell electroporation with cell positioning and impedance monitoring using micro electrode array

This paper reports a novel microarray chip for in-situ, real-time and selective electroporation on individual cells integrated with cell positioning and impedance monitoring. An array of quadrupole-electrode units (termed positioning electrodes) and pairs of planar center electrodes located at the centers of each quadrupole-electrode unit were fabricated on the chip. The positioning electrodes ...

A 3D Integrated Circuit for Sensing Biological Nanoparticles

We have designed a dielectrophoresis-based lab-on-chip using MIT Lincoln Labs 0.18 μm 3D integrated circuit technology. Dielectrophoresis is the phenomena where electric fields are used to control the movement of particles in a fluid. The topmost tier of a three chip stack (figure 1) is used to create a microfluidic trench and dielectrophoresis electrode array. During the 3D fabrication process...
